PROTEIN PHOSPHORYLATION ANALYSIS
The human genome encoded 518 protein kinases and 65 protein phosphatases which control reversible phosphorylation of proteins.
However, only about 0.05% of protein phosphorylation occurs on Tyrosine residues in mammals. Thus profiling Tyrosyl phosphorylation appears very challenging without enrichment of phosphoproteins or phosphopeptides.
Immunoprecipitation, immobilized metal affinity chromatography, methylation of peptides, nanoflow reversed phase were combined with mass spectroscopy to detect theses modified peptides in the mixture.
